In terms of Seinfeld...I'm not sure if it's my favorite one, but the movie spoof scene that always stands out the most to me is their JFK one, made even funnier by the fact that Wayne Knight was in both the spoof scene and the actual one in JFK. "That is one magic loogie."

I don't know if this fits your parameters necessarily, but I'd say the most effective jumpscare I've ever seen might be in The Conversation, a non-horror movie. Because you are not expecting it at all, and it definitely got me.
